Cdc42 localized in the CatSper signaling complex regulates cAMP?dependent pathways in mouse sperm

چکیده

Sperm acquire the ability to fertilize in a process called capacitation and undergo hyperactivation, change motility pattern, which depends on Ca2+ transport by CatSper channels. is essential for fertilization it subjected complex regulation that not fully understood. Here, we report similar CatSper, Cdc42 distribution principal piece confined four linear domains this localization disrupted CatSper1-null sperm. inhibition impaired activity other Ca2+-dependent downstream events resulting severe compromise of sperm fertilizing potential. We also demonstrate function modulating cAMP production soluble adenylate cyclase (sAC), providing new regulatory mechanism stimulation cAMP-dependent pathway. These results reveal broad mechanistic insight into mammalian sperm, matter critical importance male infertility as well contraception.

The CatSper channel controls chemosensation in sea urchin sperm.

Sperm guidance is controlled by chemical and physical cues. In many species, Ca(2+) bursts in the flagellum govern navigation to the egg. In Arbacia punctulata, a model system of sperm chemotaxis, a cGMP signaling pathway controls these Ca(2+) bursts. The underlying Ca(2+) channel and its mechanisms of activation are unknown. Here, we identify CatSper Ca(2+) channels in the flagellum of A. punc...

phylogeography and genetic diversity of the lesser mouse- eared bat (myotis blythii) in iran

in current study, 63 samples of bat populations collected from differ regions were used for evaluating the geographic variations. twenty cranial and dental characters for traditional morphometric and landmarks method on the ventral, dorsal skull and mandible for geometry morphometric studies were used. statistical analyses of traditional morphometric and geometry morphometric data indicated low...

The CatSper channel: a polymodal chemosensor in human sperm.

The sperm-specific CatSper channel controls the intracellular Ca(2+) concentration ([Ca(2+)](i)) and, thereby, the swimming behaviour of sperm. In humans, CatSper is directly activated by progesterone and prostaglandins-female factors that stimulate Ca(2+) influx. Other factors including neurotransmitters, chemokines, and odorants also affect sperm function by changing [Ca(2+)](i). Several liga...

LIS1 Regulates Osteoclastogenesis through Modulation of M-SCF and RANKL Signaling Pathways and CDC42

We have previously reported that depletion of LIS1, a key regulator of microtubules and cytoplasmic dynein motor complex, in osteoclast precursor cells by shRNAs attenuates osteoclastogenesis in vitro. However, the underlying mechanisms remain unclear. In this study, we show that conditional deletion of LIS1 in osteoclast progenitors in mice led to increased bone mass and decreased osteoclast n...

The Role of miRNA Dysregulation in Thyroid Cancer Development by Targeting the Main Signaling Pathways

Thyroid cancer is one of the most common malignancies of endocrine glands, causing carcinomas, such as papillary, follicular, medullary, and anaplastic thyroid carcinomas. Due to the significance of thyroid carcinomas, identification of the main signaling pathways and  the affecting mutations has been considered by researchers. Further studies on the dysregulation of oncogenes in signaling path...
